The clinical response to chemotherapy regimen of cisplatin and paclitaxel in lung adenocarcinoma is frequently limited in magnitude and duration due to drug resistance. Recently, epithelial-mesenchymal transition (EMT) was associated with drug resistance. However, the underlying mechanisms remain elusive. To discover mechanisms of resistance, we developed the epithelial-like and mesenchymal-like A549 cell lines (lung adenocarcinoma cell line), and have their lncRNA expressions profiled and compared. Results provide insight into the molecular mechanisms underlying chemotherapy-resistance. According to our prior report, A549 cell presented the mesenchymal phenotype on the fibronectin-coated dish. Knockdown of Caspase-8 or c-Src impaired the process of EMT to show the epithelial-like characteristics. Hence, we stably transfected control shRNA, Caspase-8 shRNA and c-Src shRNA into A549 cell, A549+Control-meshenchymal-like A549, A549+Casp8 shRNA and A549+Src shRNA-epithelial-like A549. We designed this experiment to screen for lncRNA which is required for chemotherapy-resistance in the mesenchymal-like A549 cell.
