N4-acetylcytidine (ac4C) is an evolutionarily conserved chemical modification of mRNA, catalyzed by the writer protein N-acetyltransferase 10 (NAT10), which has emerged as a key regulator of mRNA stability and translation efficiency. This epitranscriptomic modification is implicated in various cellular processes and cancer progression. THUMP domain-containing protein 1 (THUMPD1) is known to function as an essential activator and cofactor of NAT10 in the establishment of ac4C modifications on tRNA. However, whether THUMPD1 also regulates ac4C deposition on mRNA, and its potential role and mechanistic contribution to cancer biology, particularly in LUAD, remains largely unknown.
