Bone marrow, spleen and lymph node Tregs; WT anti-CD25 long-term hematopoietic stem cells

Regulatory T cells (Tregs) are a subpopulation of T cells critical for maintaining bone marrow homeostasis and regulate stromal cell function via IL-10 Live CD4+ CD8α− Foxp3+ NK-1.1− (Tregs) were sorted from these tissues (bone marrow, inguinal lymph node, and spleen) of 6-8-week-old Foxp3GFP mice and resuspended in Buffer RLT. RNA was extracted using RNAeasy micro kit (QIAGEN, Valencia, CA), with 3 biological replicates per tissue. Live Lin−Sca-1+c-Kit+Flt3-CD150+CD48− (long-term hematopoietic stem cells) were sorted from 10-12-week-old B6 mice treated with anti-CD25 or IgG1 and resuspended in Buffer RLT. RNA was extracted using RNAeasy micro kit (QIAGEN, Valencia, CA), with 4 biological replicates per treatment condition.
