CITE-Seq of CD3+ T-cells surviving CD5.28.z CART cytotoxicity

The goal of the study was to investigate the mechanism of healthy T-cell resistance to CD5.CART-directed cytotoxicity when T-cell tumors are eliminated. CD19.28.z CAR T-cells or CD5.28z CAR T-cells were co-cultured with either CD3+ T-cells freshly isolated from PBMCs or T-cell tumors (CCRF-CEM [T-ALL], Hut-78 [T-LBL]) for 36 hours. Following co-culture and eradication of tumor cell targets, residual surviving CD3+ T-cell targets were FACS sorted and subjected to single-cell CITE-Sequencing Analysis.
