KDM1A/LSD1 inhibition enhances chemotherapy response in ovarian cancer

We examined global transcriptional changes in SKOV3-control and KDM1A knockdown (KD) cells that are treated with vehicle or cisplatin using RNA-sequencing. SKOV3-control and SKOV3-KDM1A knockdown cells were treated with vehicle or cisplatin (2.5 µM) for 72 hours. Total RNA was isolated using RNeasy mini kit according to the manufacturer's instructions (Qiagen, Valencia, CA). Illumina TruSeq RNA Sample Preparation was performed following the manufacturer's protocol. Samples were run on an Illumina HiSeq 3000 in duplicate. The combined raw reads were aligned to UCSC hg19 by Tophat2. Genes were annotated and quantified by the HTSeq-DESeq pipeline.
