We targeted multiple freshwater species across taxa using multigene eDNA metabarcoding to identify multiple species across taxa along a longitudinal gradient in watersheds to understand spatial variation and species patterns in a freshwater biodiversity assessment in four neighboring streams in the Trask River watershed, Tillamook basin, coastal Oregon. Specifically, we evaluate the spatial heterogeneity of various freshwater species across the transition from below to above the distribution of fish in streams to determine how they are distributed around this boundary and to understand more generally about the distribution patterns of freshwater species, especially if we can identify rare and cryptic species. We have reads from primers targeting fishes, amphibians, crayfishes, freshwater insects, bivalves, mammals, fungi, and forest and animal pathogens.
