aSMA identifies osteochondroprogenitors in the periosteum involved in fracture callus formation. Cell surface marker expression, histological analysis, and lineage tracing data indicate that this is a heterogeneous population that contains a subset of self-renewing stem or progenitor cells. The goal of this study was to evaluate the heterogeneity of the initial cell population labeled using plate-based single cell RNAseq, and see if that changed following up to 13 weeks of chase. Periosteal cells were freshly isolated from mice and cell sorts performed directly into a 384-well plate. Animals received 2 doses of tamoxifen (75mg/kg ip) 24h apart, cells were isolated 2 days, 6 weeks, and 13 weeks later. 13 week cohort (H2B-GFP mice) received doxycycline in their diet from P11 for 5 weeks. One sort was performed (all done on the same day) for each cell type (192 cells for 2 days and 13 weeks, 216 cells for 6 weeks)
