Functional relevance of specific C/EBPD-binding sites within the S100A8 and S100A9 promotor regions was revealed by chromatin-immunoprecipitation assays and targeted mutation of DNA response elements. Using ATAC-sequencing and analysis of histone marks it could be shown that C/EBPD increases chromatin accessibility on S100A8 and S100A9 promoter regions which includes a so far unknown link of C/EBPD transcriptional activation to histone demethylase JMJD3 activity. ATAC-seq was performed to examine Chromatin accessibility in ER-Hoxb8 monocytes .
