Latrophilin 2 (ADGRL2), an adhesion GPCR, mediates signaling through its intracellular loop 3 via a unique QQQ binding patch within the Ga13 GTPase domain, playing a critical role in the regulation of normal epidermal differentiation. Overall design: Cas9 CRISPR libraries targeting GPCRs and controls were infected via lentivirus to primary keratinocytes (two biological replicates), which were then seeded for differentiation. Differentiating cells were sorted by FACS for the differentiation marker KRT10, splitting high, medium and low KRT10 populations. The fastq files are Cas9 guide sequences either from cells infected with lentivirus or plasmid DNA. In addition, libraries were generated from progenitor and differentiated cells without sorting, leading to five samples per biological replicate and a total of ten samples. The plasmid library used to generate the lentivirus was also sequenced.
