We analysed the transcriptomic gene signature of day 3 CD8+ T cell effectors in the absence of NR4A3 to determine the impact of this transcription factor on early transcriptomic programming of CD8 T cell memory and function Overall design: We compared the transcriptome of Nr4a3 deficient and sufficient CD8+ T (OT-I) cells effectors at day 3 post Listeria monocytogenes infection (strain that express OVA peptide). These effectors were generated by an adoptive transfer of Nr4a3 deficient or sufficient OT-I (CD8 CD45.2+) into a congenic B6.SJL (CD45.1+) that was then infected with a sublethal dose of Lm-OVA. The viable activated (CD44hi) cells were sorted directly in TRIzol reagent and used for RNA extraction.
