Skeletal muscle has abilities to regenerate after injuries and adapt to mechanical loadings. Compared with the regenerative processes, the cellular networks in the loaded muscles have not been well-explored. In this analyses, we did single-cell RNA sequencing to unveil the gene expression signature of individual cells and the cellular networks in surgically overloaded muscle. One side of plantaris muscles of 5 male C57BL/6J mice (12 week-old) were subjected to tenotomy and the contralateral plantaris muscles were used as the sham control. Four days after tenotomy, the plantaris muscles were pooled in each group (sham or tenotomy) digested using 0.2% collagenase type II for 90 min. After the dilution, filtration, and centrifugation of the digested solution , the mononuclear cells were stained with Calcein AM for 5 min. Sorted Calcein AM+PI- living cells were used for scRNA-seq analyses.
