This study investigated the effects of dietary sulfur amino acid restriction (SAAR) on the mouse liver transcriptome. We fed wild-type (C57Bl/6J) male mice either a control (Ctrl) or SAAR diet on either a regular-fat (RF) or high-fat HF) background for seven days. After seven days, liver samples were collected, among other tissues, and processed for RNA-sequencing. We then analyzed the changes in the transcriptome, comparing the two different Ctrl vs. SAAR comparisons against each other to identify how HF might impact the effect of SAAR on the hepatic transcriptome. Additionally, this project also involved generating proteomics data which we used to compare the similarities and differences between transcriptomic and proteomic changes in response to the different dietary treatments. We find that SAAR, independent of diet, produces a robust realignment of the transcriptome after seven days of SAAR. In contrast, proteome-wide changes were less well-aligned. Overall design: Livers from a total of 12 male mice were processed for RNA-sequencing after 7 days of consuming either a control (ctrl) of sulfur amino acid restricted (SAAR) diet on either a regularfat (RF) or high-fat (HF) background.
