To investigate mechanisms of drug resistance, we use either drug-selections or CRISPR/Cas9 editing to generate Plasmodium falciparum parasites that possess one or more mutations in their genome. In the case of drug selections, parasites grown in vitro are exposed to increasing concentrations of compound until a shift in drug susceptibility is observed. In the case of CRISPR-edited lines, the parasite is transfected with the Cas9 plasmid along with an appropriate donor template to engineer specific changes in the genome.
