RNA-Seq of four types of mouse immune samples including those of the spleen (SP), bone marrow (BM), lymph node (LN), and peripheral blood mononuclear cell (PBMC)

We performed RNA-seq experiments on a total of 12 mouse immune organs, including spleen (SP), bone marrow (BM), lymph node (LN) and Peripheral blood mononuclear cell (PBMC). Briefly, RNA-Seq libraries were constructed after rRNA depletion using a NEBNext rRNA Depletion Kit (Human/Mouse/Rat) (NEB). The E6310L NEBNext Ultra RNA Library Prep Kit for Illumina (NEB, E7530) (NEB) was used according to the manufacturer's instructions and the cDNAs were sequenced with the Hiseq X10 platform(Illumina)本研究共计对12个小鼠免疫器官开展RNA测序（RNA-seq）实验，所涉样品包括脾脏（SP）、骨髓（BM）、淋巴结（LN）及外周血单个核细胞（PBMC）。简言之，本研究先采用NEBNext® 核糖体RNA（rRNA）去除试剂盒（Human/Mouse/Rat，NEB）完成核糖体RNA去除步骤，随后构建RNA测序文库。本研究严格遵循制造商提供的操作手册，使用NEBNext® Ultra RNA文库制备试剂盒（适配Illumina测序平台，货号E6310L，NEB；货号E7530，NEB）完成cDNA合成，并依托Illumina HiSeq X10测序平台对合成的cDNA进行测序。
