IS-free E. coli strain MDS42 was transformed with plasmid pM-RK and propagated for 70 generations, with samples taken for whole genome sequencing at generations 10 and 70. Data from these samples were used to test for accumulation of insertions or novel variants during propagation as part of a broader study to evaluate mobility of transposable elements in holdings of plasmid repositories.
