This study developed a new approach, Kingcreate-Quantification (KCQ) system, for quantitative analysis of microbes in clinical specimens by mNGS, which co-sequenced DNA extracted from the specimens with synthetic dsDNA molecules used as Internal-Standard (IS), and allowed the conversion of microbial reads into their copy numbers based on the IS reads by mathematical model.
