The goal of this study is to increase the specificity of an allosteric transcription factor LasR. A combinatorial saturation mutagenesis library was introduced to LasR L125-L130 region. We then performed the pooled sort-seq approach to quantify corresponding sensor's response to cognate and noncognate signals.
