OP50 bacterial strains, commonly utilized as a food source for Caenorhabditis elegans , were subjected to mutagenesis using ethylmethanesulfonate to create random mutants. These mutants were followed by adaptive laboratory evolution for approximately 500 generations in presence of 2.5 mM paraquat. Overnight cultures of six individual clones of evolved bacteria (OP50PQE) and a single clone of wild-type bacteria (OP50) were incubated at 37 degree celsius . Genomic DNA was then isolated using the Qiagen DNeasy kit. The purified DNA samples were subsequently sent to Novogene Corporation Inc. for sequencing on the Illumina HWI-ST1276 instrument.
