SmAA10A-Cu(I) was generated via reduction of the Cu(II) congener with 50 molar equivalents of ascorbic acid in 50 mM MES pH 6.5 buffer. The protein was subsequently purified by size exclusion chromatography. Cu K-alpha HERFD-XANES was collected at ID26 configured with Si(311) double crystal monochromator and five Si(444) analyzer crystals set at the maximum of the Ka1 emission feature. Sample temperature was held at 20 K.
