Description of the project Inflammation-induced intestinal barrier dysfunction is a key feature of several gastrointestinal disorders. Bovine whey contains bioactive components with potential barrier-protective and immunomodulatory properties, although its activity may depend on fraction composition. In this study, the effects of whole bovine whey (BWW) and skimmed bovine whey (BSW) were evaluated in a differentiated Caco-2/TC7:HT29-MTX co-culture model exposed to basolateral TNF-α + IFN-γ (50 ng/mL, 24 h). Cytokine treatment significantly reduced transepithelial electrical resistance (TEER), downregulated CLDN1 and CLDN3, increased IL-1β expression, and decreased MUC2 expression, whereas FITC-dextran permeability remained unchanged. Both whey fractions restored TEER to values comparable to control and also increased TEER under basal conditions. BWW attenuated cytokine-induced IL-1β upregulation, whereas BSW increased MUC2 expression under both basal and inflammatory conditions. These results indicate that BWW and BSW improve epithelial barrier function under inflammatory conditions through distinct functional activities.
