We compared the performance of droplet digital PCR (ddPCR) and high-throughput sequencing (HTS) for the quantification of cyanobacterial target genes in sediments from two experimental (227 and 223) and two reference (224 and 442) lakes from ELA and tested the hypothesis that the two techniques provide similar information on temporal trends. The outputs from ddPCR and metabarcoding approaches, such as HTS, has not been assessed over timescales of decades using sedimentary archives. Additionally, the coherence between these two methods using samples with higher levels of PCR inhibitors in comparison to samples with less inhibition, has also not been described.
