Cellular senescence has been shown to contribute to deliterious cancer phenotyores through modulation of the tumor microenvironment through the SASP. Senescence is induced in ovarian cancer by chemotherapy. Understanding how the SASP modulates gene expression on cancer may lead to novel targets to exploit to increase chemotherapeutic efficacy. Overall design: To investigate the role of SASP on gene expresssion, fresh media and conditioned media was collected from vehicle treated proliferative cells or cisplatin therapy-induced senescent cells and used to to culture Ovcar8 human ovarian cancer cells in ultra-low attachment conditions. As the non-protein fraction of the SASP is understudied, samples were included with boiled conditioned media or conditioned media fractionated into greater the 3 kDa or less than 3 kDa components. All samples were done in triplicate.
