TAp73 functions through posttranscriptional codes in brain multiciliogenesis

Although being essential to respiratory and reproductive tracts multiciliogenesis, TAp73 is dispensable for multiciliogenesis in the ventricles. TAp73 KO is accompanied by dramatic changes in ciliogenic microRNAs miR34bc and miR449 family members, suggestin TAp73 functions partially thorugh posttranscriptional nodes in brain ciliogenesis. Total RNA from WT and TAp73 KO lateral ventricles/CP were isolated by phenol/chloroform method.
尽管TAp73对呼吸道与生殖道的多纤毛发生至关重要,但它对于脑室中的多纤毛发生并非必需。TAp73基因敲除(Knock Out, KO)会伴随纤毛发生相关微小RNA(microRNA, miRNA)miR34bc及miR449家族成员的显著变化,这提示TAp73在脑纤毛发生过程中部分通过转录后调控节点发挥功能。采用苯酚-氯仿法提取了野生型(Wild Type, WT)与TAp73基因敲除小鼠侧脑室/CP中的总RNA。
