We previously demonstrated that OK cells cultured under orbital shear stress have morphological and functional features that more closely resemble the proximal tubule in vivo. This study was designed to identify progressive transcriptional changes with increasing exposure time to shear stress and to compare the transcriptome of cells cultured under shear stress vs static conditions. OK cells plated on permeable supports were transferred to an orbital shaker the following day (t=0) and cultured up to 96h. Filters were harvested at 12, 48, and 96h for RNASeq analysis. Parallel filters maintained under static conditions were harvested at 0, 48, and 96h.
