Analysis of TCR repertoire in WT and Rras2−/−mice

A consequence of enhanced negative selection in Rras2−/−mice might manifest in a skewed TCR repertoire resulting in the generation of lower frequency of autoreactive T cell clones. To study changes in the TCR repertoire, naïve CD4+ T cells from non-immunized WT and Rras2−/−mice were selected, and the TRAV- and TRBV-encoding mRNAs were sequenced after unbiased PCR expansion.
