We take advantage of in vitro-derived motor neurons as a paradigm to investigate the cellular level during motor neuron differentiation. In this study, to uncover the abundantly expressed gene in the development of spinal motor neuron, we characterize transcriptomic signatures of HuES (human embryonic stem cell) derived motor neuron . Overall design: To investigate motor neuron maturation in a human context, we used HuES lines to differentiate into spinal motor neurons and performed strand-specific RNA-seq at indicated time points. Specifically, we profile motor neuron transcriptome at Day 8, Day 16, and Day 31, referring to motor neuron progenitor (pMN), postmitotic motor neuron, and mature motor neuron.
