The sequencing of the COI gene marker to analyse the communities of aquatic oligochaetes (metabarcoding) necessitate further testing. We tested the accuracy of the Illumina Miseq technology to recover the composition and abundance of oligochaete community by comparing the sequence data obtained for mixed oligochaetes samples with the data expected from the incorported specimens. The sequence of each incorporated specimen was obtained using from Sanger sequencing of single individuals.
