We cloned GFP-2A-Puromycin fragment in-frame in pCAGGS-hsDicer vector (Addgene Plasmid #41584) as DICER1 expressing vector and used pCDH513b (GFP) as control construct were utilized for transfection in MSCs. MSCs were treated with puromycin (2ug/ml) for 4 days and RNA extracted with TRIzol and RNA sequencing performed after rRNA depletion with NovaSeq6000.
