RNA sequencing of CD103+ cDC1 from melanoma tumors

RNA sequencing was used to characterize in situ tumor infiltrating conventional type 1 dendritic cells (cDC1) in BRAFV600E melanoma tumors transplanted into C57BL/6 wildtype mice. Our analysis shows PGE2-dependent differences in the gene expression profile of intratumoral cDC1. C57BL6 WT mice were inoculated with 2x10E6 PGE2-producing control BRAFV600E or PGE2-deficient Ptgs1/Ptgs2 BRAFV600E melanoma cells. After 6 days, cDC1 were sorted from tumors and replicates of 10 cells were sequenced using SMART-Seq3.
