In order to study if splicing of terminal introns is coupled to 3 prime end processing by cleavage and polyadenylation (CPA) of mRNAs in S. cerevisiae. Direct RNA sequencing was performed using Nanopore. RNAs were sequenced from cells that were inactivated for the essential splicing factor Snp1 (U170K homologue), or one of the CPA factors Ysh1 (CPSF73 homologue) and Rna15. Overall our results argue against a broad coupling of splicing to CPA in S.cerevisiae but show that efficient CPA mediated transcription termination is critical for splicing fidelity and efficiency of adjacent genes in a compact genome.
