Effect of Gut microbiome in the efficacy on the academic CAR-T cell ARI-0002h in multiple myeloma

Multiple myeloma is a type of blood cancer that remains incurable despite advancements in immunotherapies like CAR-T cell therapy. This study focuses on how gut microbiota may influence the effectiveness of BCMA-directed CAR-T therapy (ARI0002h) in multiple myeloma patients. Researchers analyzed stool and blood samples from patients, comparing their gut microbiome composition to healthy individuals. Using 16S rRNA sequencing and metabolomic profiling, they identified specific bacteria and metabolites linked to CAR-T cell persistence, memory, and clinical outcomes. These microbial profiles were incorporated into predictive models, including one that identified patients likely to progress from a partial to complete response by day 100 after therapy.
