To identify potential mechanisms by which EMT tumour cells are resistant to chemotherapy, we assessed the expression of putative mediators of resistance to therapy in Epcam+ and Epcam- tumour cells from skin squamous cell carcinoma. To determine how RhoJ impacts the transcriptional landscape of Epcam- tumour cells, we performed RNA seq of sh-RNA RhoJ knock-down Epcam- tumour cells. Comparative gene expression profiling analysis of RNA-seq data for FACS isolated Epcam+ or Epcam- YFP+ tumour cells from Lgr5CreER/KRasG12D/p53cKO/ Rosa-YFP mice; Comparative gene expression profiling analysis of RNA-seq data for Epcam- cells grown in culture and transduced with RhoJ sh-RNA or control sh-RNA. 2 cell lines were studied in each condition.
