Allergic and irritant contact dermatitis can be challenging to distinguish. We used single cell RNA sequencing (scRNA-seq) and insterstitial fluid proteomic analysis to compare suction blister biopsy skin samples from human volunteers with induced allergic and irritant contact dermatitis reactions. Overall design: Volunteers were first sensitized to the allergen, squaric acid dibutyl ester (SADBE). Skin reactions were induced by 2-day application of individual patches containing SADBE, an acetone vehicle control, and the irritant sodium lauryl sulfate (SLS). Skin reactions as well as a nonlesional control site were sampled by suction blister biopsy. A 4-day allergic reaction was also sampled by 2-day allergen patch application followed by 2 days of rest before the suction blister biopsy. Suction blister samples were collected by aspirating fluid from the blister. Samples were centrifuged to pellet cells, which were analyzed by scRNA-seq.
