This study conducted metatranscriptomic sequencing of soil samples collected from natural habitats of Tricholoma matsutake, a symbiotic ectomycorrhizal fungus. Total RNA was extracted from soil and sequenced using the Illumina NovaSeq 6000 platform following rRNA depletion with the Illumina Stranded Total RNA Prep with Ribo-Zero Plus protocol. The goal was to investigate active microbial communities and gene expression profiles in soil zones with high T. matsutake presence compared to adjacent areas with little or no fungal colonization. The resulting data provide insights into the functional dynamics of soil microbiota associated with T. matsutake ecology.
