The strain was isolated from honeybee residing White clover, flower (WCG5), Galtee farm located Co. Tipperary Ireland, on Fructose Yeast Peptone (FYP) agar. The strain was isolated from fructose-rich environments and is likely to utilize fructose more efficiently, and can produce mannitol at elevated amounts with greater yield. 16S RNA showed that Fructobacillus fructosus DPC 7238 was found to possess the mdh gene. Fructose tolerance tests showed extremely high tolerance of fructose i.e. 50% by DPC 7238. We found DPC 7238 produced mannitol naturally in fermented broth. F. durionis DPC 7238 displayed high fructose utilization (9.27 g/L), high mannitol yield (0.99 g mannitol/g fructose) and greater volumetric productivities (0.46g/Lh) in FMRS. Also DPC 7238 was able to produce mannitol in fructose supplemented milk, utilizing 8.57 g/L fructose and producing 4.25 g/L of mannitol with yield of 0.50 g/g fructose. Due to its mannitol production capability in fermented broth as well as milk, we further investigated its genome profile by WGS.
