BAC library of Salvia officinalis is constructed. Positive clones containing five terpene synthases are screened out. BAC Genomic DNA is extracted and then fragmented randomly. After electrophoresis, DNA fragments of desired length are gel purified. Adapter ligation and DNA cluster preparation are performed and subjected to Hiseq sequencing.
