水酶法提山苍子核仁油的提取设备简单,提取效率高,对环境影响小,在一定程度上可以解决山苍子核仁被直接丢弃、环境污染、资源浪费等问题。实验以山苍子核仁为原料优化水酶法提油工艺,得出以下结论:(1)通过复合酶制剂筛选实验发现当中性蛋白酶与纤维素酶质量比1:1,酶添加量为3%时,核仁油提取率最高。(2)通过单因素实验分析确定水酶法提山苍子核仁油的工艺条件为液料比3mL/g、pH6、温度45 ℃,时间7h。(3)在单因素实验的基础上,利用响应面实验设计分析了酶解温度、pH、液料比三个因素及其交互作用对山苍子核仁油提取率的影响,得到了提取率与各因素之间的模型,进一步优化了水酶法提油工艺。将最佳工艺修正为液料比为3mL/g、pH为6.40、温度46 ℃,核仁油提取率可达 (86.10±0.68)%,与预测值86.38%接近。(4)对最佳工艺条件下提取的山苍子核仁油进行FT-IR及GC-MS分析:山苍子核仁油在FT-IR检测出的结构都能GC-MS中找到对应的脂肪酸。GC-MS分析脂肪酸成分发现月桂酸含量高达51.92%,不饱和脂肪酸的含量占30.73%,碳数居于10~12之间的中碳链脂肪酸占68.8%,不同提油工艺提取的山苍子核仁油油脂脂肪酸成分没有明显差异。数据量为596KB。
Aqueous enzymatic extraction of Litsea cubeba kernel oil features simple equipment, high extraction efficiency and low environmental impact, which can to a certain extent alleviate the problems of direct discarding of Litsea cubeba kernels, environmental pollution and resource waste. Using Litsea cubeba kernels as raw materials, the aqueous enzymatic oil extraction process was optimized, and the following conclusions were obtained:
(1) Through the complex enzyme preparation screening experiment, it was found that the extraction rate of kernel oil reached the maximum when the mass ratio of neutral protease to cellulase was 1:1 and the enzyme addition amount was 3%.
(2) The process conditions for aqueous enzymatic extraction of Litsea cubeba kernel oil were determined via single-factor experiments: solid-liquid ratio 3 mL/g, pH 6, temperature 45 ℃, extraction time 7 h.
(3) Based on the single-factor experiments, response surface methodology (RSM) was adopted to analyze the effects of three factors (enzymatic hydrolysis temperature, pH, solid-liquid ratio) and their interactions on the extraction rate of Litsea cubeba kernel oil. A regression model between the extraction rate and each factor was established, further optimizing the aqueous enzymatic oil extraction process. The optimal process was revised to solid-liquid ratio 3 mL/g, pH 6.40, temperature 46 ℃, with the kernel oil extraction rate reaching (86.10±0.68)%, which was close to the predicted value of 86.38%.
(4) Fourier transform infrared spectroscopy (FT-IR) and gas chromatography-mass spectrometry (GC-MS) were conducted on the Litsea cubeba kernel oil extracted under the optimal process conditions: all structures detected by FT-IR for the kernel oil have corresponding fatty acids identified via GC-MS analysis. GC-MS analysis of fatty acid components revealed that the content of lauric acid was as high as 51.92%, the proportion of unsaturated fatty acids was 30.73%, and medium-chain fatty acids with carbon chain lengths ranging from 10 to 12 accounted for 68.8%. There was no significant difference in fatty acid compositions of Litsea cubeba kernel oil extracted by different oil extraction processes. The total data size of this dataset is 596 KB.
