To identify a causative variant and effector gene responsible for a 2-fold increased risk of severe COVID-19, a multi-omics platform was deployed. A combination of ATAC-seq, ChIP-seq, Micro Capture-C and NuTi Capture-C were used to characterize a range of cell types, including epithelial, fibroblast, endothelial, immune and erythroid cells. Overall design: Micro Capture-C, ATAC-seq and ChIP-seq were deployed in epithelial, endothelial and fibroblast cells to characterize the epigenetic regulatory landscape of 3p21.31 - a locus which confers a 2-fold chance of severe COVID-19 NuTi Capture-C was used in immune, endothelial, epithelial, fibroblast and erythroid cells and can be found at GSE159867.
