This study aimed to define the transcriptional programs that govern the functional specialization of gamma delta intestinal intraepithelial lymphocyte (gamma delta IEL) subsets. The primary goals were to comprehensively compare the gene expression profiles of distinct, FACS-sorted populations: specifically, CD160+ BCL6RFP+ versus CD160- BCL6RFP- gamma delta IELs to identify a BCL6-associated signature, and to determine the transcriptional consequences of conditionally knocking out key transcription factors (Bcl6, Tcf7, or Prdm1) in CD8aa+ gamma delta IELs.
