HIV is notorious for its ability to evade the immune system and anti-viral drugs. A major way HIV does so is through rapid evolution. The goal of this study was to better understand HIV's ability to tolerate mutations. Using a high-throughput technique called deep mutational scanning, we estimated the effects of all possible single amino-acid mutations to most of HIV's envelope protein. The resulting data provide a basis for disentangling the role of inherent protein-level constraints and external selection pressures in shaping Env's evolution.
