Runx1- responsive genes in mdx muscles

RNA-seq was performed to compare expression pattern of musles taken form two mice strains- mdx and mdx/Runx1f/f, which are double KO carrying a muscle specific ablation of Runx1 using a Myf5-Cre. This comparison revealed the Runx1- responsive gene set in mdx muscles. We could cross this data with prior retrieved data from previous experiments found in this GEO quary, to pinpoint Runx1 target genes in muscle regeneration. RNA was extracted from soleus muscles of 2 months old mice, n=3,4 for mdx and mdx/Runx1f/f, respectively. Differentially expressed genes were discovered using the DeSeq2 software
本研究采用RNA测序(RNA-seq)技术,比较两种小鼠品系——mdx与mdx/Runx1f/f——的肌肉组织基因表达谱。其中,mdx/Runx1f/f为双敲除品系,通过Myf5-Cre介导实现Runx1的肌肉特异性敲除。该对比分析成功鉴定出mdx肌肉中受Runx1调控的基因集。我们可将本数据集与本次GEO(Gene Expression Omnibus,基因表达综合数据库)检索获取的既往实验数据进行整合,以精准定位肌肉再生过程中Runx1的靶基因。实验样本取自2月龄小鼠的比目鱼肌(soleus muscle),其中mdx组与mdx/Runx1f/f组的生物学重复数分别为3和4。差异表达基因通过DeSeq2软件筛选鉴定。
