Rivael v0.1.0: Clinical Trial Gap Analysis for RAS/MAPK-Pathway Cancers

Findings from Rivael, an automated pipeline that analyzes published biomedical literature to identify clinical trial gaps and unstudied therapeutic hypotheses in cancer treatment. This dataset presents results from analysis of 768 published papers on RAS/MAPK-pathway cancers. The pipeline extracted 669 biomedical entities and 797 relationships, scored 73 candidate hypotheses, and applied multi-stage quality filtering (removing on-label drugs, duplicates, and previously known connections) to surface 1 genuinely novel finding alongside 6 validated known connections that confirm the methodology works correctly. Key finding: RAS(ON) inhibitors — a novel class targeting the active GTP-bound state of KRAS — have no registered clinical trials for PDAC, non-small cell lung cancer, or AML despite strong biological rationale (8.0/10 plausibility). KRAS mutations drive ~90% of PDAC, ~30% of NSCLC, and 15-20% of AML, yet this therapeutic approach remains clinically untested across all three indications. The dataset includes full plausibility assessments, suggested trial designs, safety analyses, and supporting evidence references for all findings. Rivael is an ongoing project. Future versions will expand to additional disease areas including glioblastoma, DIPG, and rare pediatric cancers.
