Lichtheimia ramosa JMRC:FSU:6197 GDA DTT HEAT stress RNA-seq

100 ml of SUP medium were inoculated with 5x105 Lichtheimia spores and incubated for 16 h (NaCl, heat stress) or 20 h (GdA and DTT treatment). Then stress factors were added: Gda 5.5 µM from a 10 mg/ml stock in DMSO, DTT 5 mM and incubated for 2h. Heat (42°C) stress was applied for 6h. RNA extraction was performed using RNAeasy Plant kit (Quiagen). Strand-specific whole transcriptome libraries were prepared using the Ion Total RNA-Seq Kit v2.0 and sequenced on an Ion Proton Sequencer (Life Technologies USA).
