ChIP-seq for H3K9me3 was performed in A2780 (platinum sensitive) and A2780cis (platinum resistant) cells and in sensitive A2780 cells with shRNA induced knockdown of FLYWCH1. Overall design: Cells were transduced with control shRNAs and shRNAs targeting FLYWCH1. Treatment with 1 µM cPt was started 7 days after knockdown induction. After completion of the experiment (week 7) cells were sorted for shRNA expressing cells and ChIPseq for H3K9me3 performed.
