To study the role of MLL complex in transcriptional dysregulation of NSC aging, we performed ChIP-seq of MLL complex subunits (Kmt2d, Rbbp5, and Wdr5) and Cbx3 in young and aged NSCs to analyze their binding activity at promoters of age-dependent genes. Overall design: Primary NSCs were isolated from the subventricular zone of the lateral ventricles (SVZ) in the brains of young and aged mice, respectively. The NSCs were maintained in the initial proliferation medium and used in ChIP-sequencing analysis.
