Mitochondrial carrier SLC25A19 is required for respiratory chain function

In this study, we investigate how mitochondrial thiamine pyrophosphate (TPP) transporter SLC25A19 regulates mitochondrial function and cell metabolism. We performed CRISPR-cas9 to knock out SLC25A19 in K562 cells using two different sgRNAs. Two knockout cell lines, SLC25A19-sg1 and SLC25A19-sg2, were selected and further used for phenotype analysis. We also used K562 cells with Non-targeting control sgRNA as a reference. RNA was collected after 48 h of culturing, and each cell line was performed in triplicate.
