项目历时8年的研究与推广应用,选出高含油互叶白千层优株,在优株组培扦插快繁、栽培、加工利用方面开展了系统研究,针对互叶白千层产业化关键技术瓶颈,构建了从优良单株选育、组培快繁、高效栽培、高纯度4-松油醇加工应用及产品开发的完整产业化体系,技术性能可靠,可操作性、实用性强,具有极高应用价值。优良单株选择、组培产业化、高效栽培配套技术及高纯度4-松油醇提取工艺及产品应用等方面有创新,成果达到国内同类研究领先水平。成果推广应用促进了互叶白千层种植、深加工产业的快速发展。 1.高含油互叶白千层优良单株选择。选出高生物量、高含油优良单株21株,其精油质量指标(检测报告:4-松油醇含量46.0%、1,8-桉叶素含量3%)超过“茶树油”国际标准(IS04730-2004) 优株枝叶生物量增益1.83倍,含油率增益1.29倍,选择增益效果显著。 2.高含油优株组培产业化技术。研制出高含油优株组培产业化的独特培养基配方,营建了12个优株组培产业化技术体系。解决了优株组培苗褐化、黄化、死亡等问题,提高了组培效率,继代增殖倍率提高到5.3,生根率及组培苗移栽成活率均达95%以上,组培苗高含油优良性状不变异。 3.高含油采穗圃营建及扦插快繁技术。提出了优株组培苗栽植密度、截干促萌高度等采穗圃营建及组培苗嫩枝扦插快繁技术体系。扦插生根率从78%提高到97.5%,提高了快繁效率,降低苗木成本。 4.高含油优株高效栽培技术。高含油组培苗与扦插苗对比试验,提出了组培苗最佳种植密度、施肥措施、采收时间、采收方式的高效栽培及配套技术模式,平均枝叶产量达3.0吨/亩• 年,营建了组培苗高效丰产栽培示范基地1050亩。 5.一次性高得率提取高纯度4-松油醇及加工利用。采用特制精馏设备及设备钝化技术,通过除酸、添加滞留剂等工艺,优化了“茶树油”提取及加工工艺,提高了精油产量、质量。4-松油醇一次性提取率达85%,总提取率96%,纯度达99.7%,超过出口标准及福建省地方标准。以高纯度4-松油醇为主要原料,开发无公害天然除草剂、真菌清霜等药用、日用、生物制药新产品16个。 6.授权专利、制定标准、发表论文 获国家发明专利授权5个,外观设计专利4个 制定《4-松油醇(茶树油型)》福建省地方标准1个,新产品企业标准2个,发表学术论文7篇。
This 8-year research, promotion and application project selected elite strains of high-oil Melaleuca alternifolia, and carried out systematic research on tissue culture and rapid propagation, cultivation, processing and utilization of these elite strains. Aiming at the key technical bottlenecks in the industrialization of Melaleuca alternifolia, a complete industrialization system was established, covering elite individual selection, tissue culture rapid propagation, efficient cultivation, high-purity 4-terpineol processing and application, and product development. The technology has reliable performance, strong operability and practicability, and has extremely high application value. Innovations have been made in elite individual selection, tissue culture industrialization, supporting efficient cultivation technologies, high-purity 4-terpineol extraction processes and product applications, and the achievements have reached the leading level in domestic similar research. The promotion and application of the achievements have promoted the rapid development of Melaleuca alternifolia planting and deep processing industries. 1. Selection of elite high-oil Melaleuca alternifolia strains: 21 elite strains with high biomass and high oil content were selected. Their essential oil quality indicators (test report: 4-terpineol content 46.0%, 1,8-cineole content 3%) exceed the international "tea tree oil" standard (ISO 4730-2004). The biomass gain of elite strain branches and leaves was 1.83 times, and the oil content gain was 1.29 times, with significant selection gain effect. 2. Tissue culture industrialization technology for high-oil elite strains: A unique medium formula for tissue culture industrialization of high-oil elite strains was developed, and a technical system for tissue culture industrialization of 12 elite strains was established. The problems of browning, yellowing and death of elite strain tissue culture seedlings were solved, and the tissue culture efficiency was improved. The subculture multiplication rate was increased to 5.3, the rooting rate and transplanting survival rate of tissue culture seedlings both reached over 95%, and the high-oil elite traits of tissue culture seedlings did not mutate. 3. Construction of high-oil scion orchard and rapid cutting propagation technology: A technical system for scion orchard construction and tender branch cutting rapid propagation of tissue culture seedlings was proposed, including planting density of elite strain tissue culture seedlings and height of stem cutting for sprouting promotion. The cutting rooting rate was increased from 78% to 97.5%, improving rapid propagation efficiency and reducing seedling cost. 4. Efficient cultivation technology for high-oil elite strains: Through comparative experiments between tissue culture seedlings and cutting seedlings of high-oil strains, an efficient cultivation and supporting technical model was proposed, including optimal planting density, fertilization measures, harvesting time and harvesting method for tissue culture seedlings. The average branch and leaf yield reached 3.0 tons/mu·year, and a 1050-mu demonstration base for high-yield and efficient cultivation of tissue culture seedlings was established. 5. One-step high-yield extraction of high-purity 4-terpineol and processing utilization: Using special rectification equipment and equipment passivation technology, through processes such as acid removal and addition of retention agents, the "tea tree oil" extraction and processing technology was optimized, improving the yield and quality of essential oil. The one-step extraction rate of 4-terpineol reached 85%, the total extraction rate was 96%, and the purity reached 99.7%, exceeding export standards and Fujian Provincial local standards. Taking high-purity 4-terpineol as the main raw material, 16 new products including pollution-free natural herbicides, fungistatic creams and other medicinal, daily chemical and biopharmaceutical products were developed. 6. Authorized patents, standard formulation and published papers: 5 national invention patents and 4 design patents were authorized. 1 Fujian Provincial local standard "4-terpineol (tea tree oil type)" and 2 new product enterprise standards were formulated, and 7 academic papers were published.
