System level view of the vaccine induced immune response to identify perturbations at the level of gene expression in whole PBMC specifically in response to pertussis antigenic challenge in vivo (Tdap vaccine boost) as a proxy of infectious challenge, and whether this response differed in aP vs. wP primed individuals 15 years or more after the original vaccination. RNA-Seq analysis on PBMC samples collected longitudinally at baseline and following Tdap booster vaccination. For this purpose, we recruited addults primed with either aP and wP and proceeded to booster vaccination with aP. PBMCs were collected for transcriptomics at 0 days post boost (baseline) followed by 1, 3, 7 and 14 days post boost. Bulk PBMC RNA sequencing was performed for 39 donors with 5 timepoints (0, 1, 3, 7 and 14 days post boost) +1 donor with 3 timepoints (0, 1 and 3 days post boost) To determine immunological differences associated with aP vs. wP priming, we performed RNA expression profiling of the immune response in adults primed with aP vs. wP vaccine in infancy following the Tdap booster vaccination. A four part format of the samples was used internally using Sample number_DonorID_visit_Timepoint. Ex: 1_2685_1_TimePoint1 where 1 accounts for sample number in RNA sequencing plate, 2685 is the donor ID, 1 is the internal visit number where the blood was collected and Timepoint1 accounts for 0 days post boost (Baseline). Thus, Timepoint2, 3, 4 and 5 account for 1, 3, 7 and 14 days post boost.
