Purpose: to investigate transcriptomal differences between wild-type and SXR/PXR knockout mice and the impact of PCB-153 exposure to both strains to help reveal the mechanism behind the phenotype of hemolytic anemia observed in SXR knockout mice exposed to PCB-153 Wild-type and SXR/PXR knockout mice were exposed to approximately 54µg/kg/day of PCB-153 or vehicle (DMSO) perinatally until 4 weeks of age. Liver and spleen tissues sections were flash frozen then rapidly homogenized in Trizol reagent. Total RNA was isolated by chloroform extraction. RNA-seq libraries were prepared with the Illumina TruSeq stranded mRNA library prep kit. Two separate libraries were made for the liver and spleen samples and sequenced on the HiSeq4000 with 1 library per flow cell lane.
