The project focuses at adipose tissue derived mesenchymal stem cells (ADSCs) and theirpotential to improve skeletal muscle regeneration. The choice of ADSCs is supported by their availability as a source of cells for auto-transplants. These cells will be characterized in vitro and then subjected to exogenous factors improving their ability to migrate within the tissue and support muscle reconstruction. The cells of the best characteristic will be transplanted into injured muscles and their impact at the regeneration will be assessed. ADSCs will be also analyzed using advanced techniques, i.e. next generation sequencing, allowing to correlate their pro-regenerative potential with the methods of their pretreatment. As a result the protocol allowing preparation of ADSCs characterized by the best potential to support skeletal muscle regeneration will be designed. We will also propose the test permitting examination of the "usefulness" of given cells in order to use them in clinics.
